目的:建立甘肃1年生,2年生红芪和黄芪指纹图谱,对比研究红芪和黄芪指纹图谱相似度。方法:采用HPLCELSD,选用Agilent ZROBAX-SBC_(18)色谱柱(4.6 mm×250 mm,5μm),以水(A)-乙腈(B)为流动相,梯度洗脱,柱温35℃,进样体积15μL,Alltech ELSD 2000蒸发光检测器,漂移管温度110℃,载气流速3.0 L·min~(-1),分别建立1年生和2年生红芪各24批以及1年生和2年生黄芪各24批药材指纹图谱。结果:1年生,2年生红芪药材共有峰分别为8,7个,相似度分别为0.923,0.825,相同色谱峰6个,分别为毛蕊异黄酮苷,芒柄花苷,大豆皂苷Ⅰ,芒柄花素,异黄芪皂苷Ⅱ,黄芪皂苷Ⅰ;1年生,2年生黄芪药材共有峰分别为25,17个,相似度分别为0.980,0.997,相同色谱峰7个,分别为毛蕊异黄酮苷,芒柄花苷,毛蕊异黄酮,黄芪甲苷,黄芪皂苷Ⅲ,异黄芪皂苷Ⅱ,黄芪皂苷Ⅰ;1年生红芪与黄芪相似度为0.032,2年生红芪与黄芪相似度为0.023,共有色谱峰均为4个,分别为毛蕊异黄酮苷,芒柄花苷,异黄芪皂苷Ⅱ,黄芪皂苷Ⅰ。结论:在所确定的色谱条件下,1年生和2年生红芪和黄芪药材指纹图谱差异明显,可有效区分两种药材,二者共有成分少,相似度低,故红芪和黄芪不宜替代使用。
OBJECTIVE: To establish a one-year, two-year-old radix salviae miltiorrhizae and radix astragalus fingerprinting map of Gansu Province, and to compare the fingerprint similarity of Radix Astragali and Radix Astragali. Results: The common peaks of one year old and two year old Radix Astragali were 8 and 7, respectively, with similarities of 0.923 and 0.825, respectively, and 6 identical chromatographic peaks, including calycosin, monensin, soybean saponin Ⅰ, Astragalus saponin Ⅱ, Astragalus saponin Ⅰ; The common peaks of 1-year-old and 2-year-old Astragalus were 25,17 and the similarities were 0.980 and 0.997, respectively. The same chromatographic peaks were 7, The similarity of 1-year-old Radix Astragali and Astragalus was 0. 032, the similarity of 2-year-old Radix Astragali and Astragalus was 0.023, the common chromatographic peaks were 4, respectively, calycosin, monascus glycosides, different astragaloside Ⅱ, Astragaloside Ⅰ. Conclusion: The fingerprints of Radix Hedysari and Radix Astragali in 1-year-old and 2-year-old have obvious difference under the determined chromatographic conditions, which can effectively distinguish two kinds of medicinal materials, which have little common components and low similarity, therefore, it is not suitable to use Radix Astragali and Astragalus instead .
