目的:观察宫颈腺癌和正常宫颈腺体组织中高危人乳头瘤病毒(hr-HPV)(16/18)检出率和P16蛋白表达的差异,探讨hr-HPV(16/18)感染与P16蛋白表达在宫颈腺癌发生中的可能关系。方法:采用原位杂交及免疫组织化学EliVisionTMplus染色技术,分别对33例宫颈腺癌和25例正常宫颈腺体组织中hr-HPV(16/18)DNA及P16蛋白的表达进行检测。结果:宫颈腺癌hr-HPV(16/18)DNA检出率明显高于正常宫颈腺体组织(72.7%比24.0%,P<0.01),但宫颈腺癌各亚型间在hr-HPV(16/18)DNA检出率上的差异无显著性(P>0.05)。P16蛋白在宫颈腺癌中的高表达率(72.7%)显著高于正常宫颈腺体组织(无高表达病例)(P<0.01)。但P16蛋白在宫颈腺癌不同亚型间的高表达率并无明显差异(P>0.05)。P16蛋白高表达率在宫颈腺癌hr-HPV(16/18)DNA阳性病例(95.8%)显著高于阴性病例(11.1%)(P<0.01)。结论:宫颈腺癌与正常宫颈腺体组织在hr-HPV(16/18)DNA检出率及P16蛋白高表达率上存在明显差异性。在宫颈腺癌中,P16蛋白的高表达与hr-HPV DNA检出率有密切关系。
Objective: To observe the differences of HPV16 (16/18) detection rate and P16 protein expression between cervical adenocarcinoma and normal cervical gland tissue and to investigate the relationship between hr-HPV (16/18) infection and P16 Possible Relationship between Protein Expression and Cervical Adenocarcinoma. Methods: The expression of hr-HPV (16/18) DNA and P16 protein in 33 cases of cervical adenocarcinoma and 25 cases of normal cervical gland tissues were detected by in situ hybridization and immunohistochemistry EliVisionTMplus staining. Results: The detection rate of hr-HPV (16/18) DNA in cervical adenocarcinoma was significantly higher than that in normal cervical adenocarcinoma (72.7% vs. 24.0%, P <0.01) There was no significant difference in DNA detection rate (P> 0.05). The high expression rate of P16 protein in cervical adenocarcinoma (72.7%) was significantly higher than that in normal cervical gland tissues (without high expression) (P <0.01). However, there was no significant difference in the high expression rate of P16 between different subtypes of cervical adenocarcinoma (P> 0.05). The high expression rate of P16 protein in cervical adenocarcinoma hr-HPV (16/18) DNA positive cases (95.8%) was significantly higher than that in negative cases (11.1%) (P <0.01). Conclusion: There is a significant difference in the detection rate of hr-HPV (16/18) DNA and the high expression rate of P16 between cervical adenocarcinoma and normal cervical gland. In cervical adenocarcinoma, high expression of P16 protein is closely related to the detection rate of hr-HPV DNA.
