研究了小蜡树枝叶中皂甙类物质的提取工艺及其抗氧化活性。应用单因素和正交试验对浸提温度、乙醇浓度、浸提时间、料液比进行了研究,同时采用邻二氮菲体系和水杨酸体系研究了小腊树枝叶皂甙清除·OH的抗氧化活性。实验得出小蜡树枝叶中总皂甙的最佳提取条件是以80%的乙醇作提取剂,1:70的料液比,温度为85℃,提取时间为2h。在最佳浸提条件下,小腊树叶中皂甙的得率为39.270mg/g,小腊树枝中皂甙的得率为22.998mg/g。在水杨酸体系中小腊树枝、叶的皂甙对羟自由基的EC50为0.937、0.720mg/mL,二者都低于Vc(EC50=0.098mg/mL)的。在邻菲啰啉体系中小腊树叶、枝皂甙、Vc对羟自由基的清除率相当,分别为0.259、0.253、0.259mg/mL。
The extraction technology and anti-oxidative activity of saponins in leaves and leaves of waxy wax were studied. Single factor and orthogonal experiment were used to study the extraction temperature, ethanol concentration, extraction time and the ratio of solid to liquid. At the same time, the phenanthrene scavenging and OH scavenging activities were studied using phenanthroline and salicylic acid system Oxidative activity. The optimum conditions for extraction of total saponins from leaves and leaves of Siberian wax were 80% ethanol as extractant, 1: 70 ratio of solid to liquid, 85 ℃ temperature and 2h extraction time. Under the optimum extraction conditions, the yield of saponin in XiaoLai leaves was 39.270 mg / g, the yield of saponins in XiaoLong branches was 22.998 mg / g. In the salicylic acid system, the EC50 of saponins of small wax tree and leaf on hydroxyl radical was 0.937 and 0.720 mg / mL, both lower than Vc (EC50 = 0.098 mg / mL). In orthophenanthroline system, the clearance rate of hydroxyl radicals from Volatile Rhizome, Saponin and Vc was equivalent to 0.259, 0.253 and 0.259 mg / mL, respectively.
