目的 :对中性粒细胞明胶酶相关脂萼蛋白 (neutrophilgelatinase associatedlipocalin ,NGAL)基因 5′侧翼区的转录调控元件进行分段定位鉴定。方法 :以双荧光素酶报告基因检测系统检测NGAL基因 5′侧翼区 ( -14 3 1~ +84)转录调控元件与转录调控蛋白之间的相互作用。结果 :在NGAL基因 5′端 -945~ -65 8和 -65 7~ -4 172个区段发现了转录增强子元件与转录激活蛋白之间的相互作用 ,而在 -4 16~ -15 2区段则发现了转录沉默子元件与转录抑制蛋白之间的相互作用。结论 :在NGAL基因5′侧翼转录调控区可能至少存在着 2个转录增强子元件和 1个转录沉默子元件
OBJECTIVE: To segmentally identify transcriptional regulatory elements in the 5 ’flanking region of the neutrophil gelatinase associated lipocalin (NGAL) gene. Methods: The dual luciferase reporter gene detection system was used to detect the interaction between transcriptional regulatory elements and the transcription regulatory proteins in the 5 ’flanking region of NGAL gene (-14 3 1 ~ +84). Results: The interaction between transcriptional enhancer and transcriptional activator was found in -945 ~ -658 and -657 ~ -472 segments of 5 ’end of NGAL gene, while in -416 ~ -152 The segment then found the interaction between transcriptional silencer elements and transcriptional repressor proteins. CONCLUSION: There may be at least two transcriptional enhancer elements and one transcriptional silencer element in the 5 ’flanking transcriptional regulatory region of NGAL gene
