目的建立高分离度快速液相色谱(RRLC)方法,同时测定杏苏止咳口服液中橙皮苷和苦杏仁苷的含量。方法采用ZORBAX SB-C18色谱柱(4.6 mm×150 mm,1.8μm);梯度洗脱,流动相为0.2%磷酸溶液-乙腈;检测波长为207 nm;流速为1.0 mL·min-1;柱温为25℃;进样体积2.0μL。结果橙皮苷和苦杏仁苷进样量分别在0.007568~0.15136μg和0.03238~0.6476μg范围内呈良好的线性关系(r=0.9999、r=1)。平均加样回收率(n=6)分别为100.40%、99.79%,RSD分别为0.95%、0.53%。结论本方法简便可靠,在较短的时间内获得较高的分离度,溶剂消耗亦大幅减少,可用于杏苏止咳口服液的质量控制。
OBJECTIVE To establish a high resolution liquid chromatography (RRLC) method for simultaneous determination of hesperidin and amygdalin in Xingsu Zhike Oral Liquid. Methods The mobile phase was ZORBAX SB-C18 column (4.6 mm × 150 mm, 1.8 μm). The mobile phase consisted of 0.2% phosphoric acid solution-acetonitrile. The detection wavelength was 207 nm and the flow rate was 1.0 mL · min-1. 25 ° C; injection volume 2.0 μL. Results The injection of hesperidin and amygdalin showed a good linearity (r = 0.9999, r = 1) in the range of 0.007568 ~ 0.15136μg and 0.03238 ~ 0.6476μg, respectively. The average recoveries (n = 6) were 100.40% and 99.79%, respectively, with RSDs of 0.95% and 0.53%, respectively. Conclusion The method is simple and reliable, obtains higher resolution in a short period of time, and greatly reduces solvent consumption. It can be used for the quality control of Xingsu Zhike Oral Liquid.
