目的检测P-糖蛋白(P-gp)与缺氧诱导因子-1α(HIF-1α)在口腔鳞状细胞癌(OSCC)中的表达及相互关系,为临床评估OSCC化疗耐药性提供参考。方法选取2013年3月至12月在广西医科大学口腔医院住院行手术治疗的28例OSCC患者,对其手术切除标本行免疫组织化学SP法检测OSCC癌组织中HIF-1α蛋白和P-gp蛋白的表达。结果 28例OSCC癌组织中,HIF-1α蛋白阳性表达21例(75.0%),P-gp蛋白阳性表达20例(71.4%)。两者阳性表达率在患者不同性别、肿瘤TNM分期、病理分级及淋巴结转移中比较,差异无统计学意义(P均>0.05)。HIF-1α蛋白阳性表达率在不同大小肿瘤(P=0.029)、年龄(P=0.041)中存在差异;P-gp蛋白阳性表达率在不同大小肿瘤中存在差异(P=0.007)。28例OSCC组织中两者阳性表达一致者18例,列联相关性分析示,P-gp和HIF-1α在OSCC癌组织中的表达呈正相关性(φ相关系数=0.548)。结论 P-gp和HIF-1α的检测可为临床OSCC患者化疗耐药性的评估提供参考。
Objective To detect the expression of P-glycoprotein (P-gp) and hypoxia inducible factor-1α (HIF-1α) in oral squamous cell carcinoma (OSCC) and to provide a reference for clinical evaluation of drug resistance in OSCC. Methods Twenty-eight patients with OSCC surgically treated at the Hospital of Stomatology, Guangxi Medical University from March 2013 to December 2013 were enrolled in this study. Immunohistochemical SP method was used to detect the expression of HIF-1α protein and P-gp protein in OSCC tissues expression. Results The positive expression of HIF-1α protein was found in 21 cases (75.0%) and in 20 cases (71.4%) of 28 cases of OSCC. There was no significant difference between the two groups in gender, tumor TNM stage, pathological grade and lymph node metastasis (all P> 0.05). The positive rates of HIF-1α protein in different size tumors (P = 0.029) and age (P = 0.041) were different. The positive rates of P-gp protein in different size tumors were different (P = 0.007). In 28 cases of OSCC, 18 cases were consistent with the positive expression of OSCC. The correlation of P-gp and HIF-1α in OSCC was positively correlated (φ correlation coefficient = 0.548). Conclusion The detection of P-gp and HIF-1α may provide reference for the evaluation of chemoresistance in clinical OSCC patients.
