目的:研究TIG2(tazarotene induced gene-2)在皮肤鳞癌中的作用机制。方法:用免疫组化和原位杂交的方法检测正常皮肤组织,未受累组织及鳞癌皮损中TIG2蛋白和mRNA的表达变化。结果:TIG2蛋白和mRNA表达于正常组织及未受累组织的表皮全层;在鳞癌的边缘皮损中,可见TIG2表达于角质层,棘层上中部,而不表达于棘层下部及基底层;在Ⅰ~Ⅱ期鳞癌中,TIG2可轻度表达于分化较好的角化珠周围;而在Ⅲ~Ⅳ期鳞癌中无表达。在正常组织和未受累皮肤中,TIG2的表达高于鳞癌组织中的表达(P<0.01);边缘皮损基底层中的表达低于正常组织及未受累组织基底层中的表达(P<0.01);在Ⅲ~Ⅳ期鳞癌中的表达明显低于鳞癌边缘皮损中的表达(P<0.01)。结论:TIG2可能参与鳞癌的发生和发展。
Objective: To study the mechanism of TIG2 (tazarotene induced gene-2) in squamous cell carcinoma of the skin. Methods: Immunohistochemistry and in situ hybridization were used to detect the expression of TIG2 protein and mRNA in normal skin tissue, non-affected tissue and squamous cell carcinoma lesions. Results: TIG2 protein and mRNA were expressed in the whole epidermis of normal and uninvolved tissues. In the marginal lesions of squamous cell carcinoma, TIG2 was expressed in the middle of stratum corneum and stratum spinosum but not in the lower part of stratum spinosum and basal layer In stage I-II squamous cell carcinoma, TIG2 was slightly expressed around well-differentiated keratinocytes, but not in stage III-IV squamous cell carcinomas. The expression of TIG2 in normal and non-affected skin was higher than that in squamous cell carcinoma (P <0.01). The expression of TIG2 in basal layer of marginal lesions was lower than that in normal and non-affected tissues (P < 0.01). The expression in stage Ⅲ-Ⅳ squamous cell carcinoma was significantly lower than that in marginal lesions of squamous cell carcinoma (P <0.01). Conclusion: TIG2 may be involved in the occurrence and development of squamous cell carcinoma.
