目的:观察卡维地洛对阿霉素诱导的扩张型心肌病(dilated cardiomyopathy,DCM)大鼠心室肌缝隙连接蛋白43(connexin 43,Cx43)表达的影响。方法:应用多柔比星腹腔注射建立DCM大鼠模型;模型组大鼠按照不同的给药方法随机分为卡维地洛组和模型对照组,卡维地洛组应用卡维地洛加双蒸馏水灌胃,模型对照组和正常对照组用定量的双蒸馏水灌胃,三组均1次/d,灌胃4周。12周末超声测量各组大鼠心脏左心室射血分数(LVEF),逆转录聚合酶链反应法检测Cx43 mRNA,采用免疫组化方法观察Cx43分布。结果:与模型对照组比较,卡维地洛组大鼠心肌Cx43mRNA、Cx43平均灰度值、LVEF及均明显增高,卡维地洛组较模型对照组心肌Cx43的分布紊乱有改善,差异均有统计学意义。结论:卡维地洛能改善DCM大鼠心肌Cx43表达和分布,并能改善其心功能。
Objective: To observe the effect of carvedilol on the expression of connexin 43 (Cx43) in doxorubicin-induced dilated cardiomyopathy (DCM) rats. Methods: The rat model of DCM was established by intraperitoneal injection of doxorubicin. The model rats were randomly divided into carvedilol group and model control group according to different administration methods. Carvedilol group was treated with carvedilol plus bis The rats in the model group and the normal control group were given gavage with double distilled water. The three groups were given once daily for 4 weeks. The left ventricular ejection fraction (LVEF) was measured by ultrasound at 12weeks, Cx43 mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR), and Cx43 distribution was observed by immunohistochemistry. Results: Compared with the model control group, the myocardial Cx43mRNA, Cx43 mean gray value and LVEF were significantly increased in the carvedilol group, while the myocardial Cx43 distribution in the carvedilol group was improved more than that in the model control group Statistical significance. Conclusion: Carvedilol can improve the myocardial Cx43 expression and distribution in DCM rats and improve its cardiac function.
