目的研究大鼠脑缺血再灌注后水通道蛋白-4(Aquaporin-4,AQP4)的表达变化。方法采用栓线法制作大鼠大脑中动脉局灶性缺血再灌注模型,用免疫组化检测大鼠脑内AQP4的动态变化。结果大鼠脑AQP4蛋白表达与缺血时间、再灌注时间均有相关性(P值分别为P<0.001,P<0.05)。结论缺血时间与再灌注时间对AQP4在鼠脑中的表达均有影响,且存在交互作用。缺血时间越短进行再灌注,脑内AQP4表达水平越低,而缺血6h后实施再灌注则会导致AQP4急剧增高。
Objective To investigate the expression of aquaporin-4 (AQP4) after cerebral ischemia-reperfusion in rats. Methods The middle cerebral artery occlusion (MCAO) model of focal ischemia-reperfusion was established by the method of thread occlusion. The dynamic changes of AQP4 in rat brain were detected by immunohistochemistry. Results The expression of AQP4 in rat brain was correlated with ischemia time and reperfusion time (P <0.001, P <0.05, respectively). Conclusion The ischemic time and reperfusion time have an effect on the expression of AQP4 in rat brain and there is interaction. Shorter ischemic reperfusion, brain expression of AQP4 lower, and 6h after ischemia reperfusion will lead to a sharp increase in AQP4.
