目的:研究姬松茸中分离纯化的低分子量多糖(ABP-AW1)促进卵清白蛋白(ovalbumin,OVA)抗原诱导小鼠脾淋巴细胞亚群效应细胞的应答格局的影响。方法:ABP-AW1和OVA皮下两点免疫ICR小鼠,于0,15 d各免疫一次,第28天分离培养脾淋巴细胞,MTT法测定脾淋巴细胞增殖,FCM(flow cytometry)分析免疫鼠脾中T淋巴细胞亚群CD4+T和CD8+T的百分含量;免疫鼠脾淋巴细胞体外给予同种抗原再刺激24 h,分析ABP-AW1对脾CD69+CD4+T淋巴细胞的表达及胞内因子IFN-γ+CD4+T的调节作用。结果:ABP-AW1能增强OVA诱导的Th淋巴细胞亚群CD4+T细胞,CTL淋巴细胞亚群CD8+T细胞增殖;ABP-AW1也能增进CD4+T细胞早期活化标志CD69+百分比增加,诱导CD4+T胞内Th1型细胞因子IFN-γ分泌水平增多。结论:ABP-AW1佐剂上调脾脏中淋巴细胞亚群的增殖,特别是Th1细胞的募集和活化,研究结果为设计理想的免疫佐剂提供了基础研究数据资料。
OBJECTIVE: To study the effect of low molecular weight polysaccharide (ABP-AW1) isolated from Agaricus blazei Murill on the response patterns of spleen lymphocyte subsets induced by ovalbumin (OVA) antigen in mice. Methods: ICR mice were immunized subcutaneously with ABP-AW1 and OVA for 0, 15 days. Spleen lymphocytes were isolated and cultured on the 28th day. Splenic lymphocyte proliferation was measured by MTT assay. FCM (flow cytometry) T lymphocyte subsets CD4 + T and CD8 + T percentage of splenic lymphocytes were stimulated with alloantigen in vitro for 24 h. The expression of ABP-AW1 in splenic CD69 + CD4 + T lymphocytes and cells Regulatory effect of IFN-γ + CD4 + T. Results: ABP-AW1 could enhance the proliferation of CD4 + T lymphocytes and CTL lymphocyte subsets CD8 + T cells induced by OVA. ABP-AW1 could also increase the percentage of early activation markers CD69 + of CD4 + T cells and induce CD4 + T intracellular Th1-type cytokine IFN-γ secretion increased. CONCLUSION: ABP-AW1 adjuvant up-regulates the proliferation of lymphocyte subsets in spleen, especially the recruitment and activation of Th1 cells. The results provide the basic research data for the design of ideal immune adjuvant.
