将人精液分为两组,一组直接用于制备精子单倍染色体,一组保存于4~8℃的HT卵黄缓冲液中48小时,再用于制备精子单倍染色体。比较两组的可分析染色体获得率与自然畸变率,了解HT卵黄缓冲液是否会造成自然畸变率的上升和可分析染色体获得率的下降。对35份样品试验与观察得到两组间可分析染色体获得率比较结果,X2=0.159,P>0.05;自然畸变率两组比较结果,X2=0.074,P>0.05。结果表明两组间在这两个率比较中无统计学差异,HT卵黄缓冲液可应用于精子在冷环境中短期贮藏,并不具有致染色体突变的效应。
Human sperm were divided into two groups. One group was used to prepare sperm single chromosomes directly. One group was stored in HT egg yolk buffer at 4 ~ 8 ℃ for 48 hours and then used to prepare sperm haploid chromosomes. Comparing the analyzable chromosomal acquisition rate and the natural aberration rate in both groups, it is possible to know whether HT yolk buffer causes a rise in the rate of natural aberration and an analysis of the reduction in chromosomal yield. The results of the comparison between the two groups in the test and observation of 35 samples showed that X2 = 0.159, P> 0.05; the natural distortion rate was X2 = 0.074, P> 0.05 . The results showed no significant difference between the two groups in the two rates, HT yolk buffer can be applied to short-term sperm storage in cold environment, does not have the effect of chromosomal mutation.
