目的探讨中华眼镜蛇毒C组分(FC)对胶质瘤U251细胞株的抑制作用及原癌基因Fas/FasL表达的影响。方法用MTT比色法检测不同浓度FC对U25l细胞的增殖抑制,免疫组织化学和RT-PCR技术观察Fas/FasL表达的变化。结果随着FC浓度的增加,对U251细胞的增殖抑制作用逐渐增强,呈剂量依赖性。免疫组化和RT-PCR结果显示,随着FC作用剂量的增加,Fas/FasL表达无明显改变。结论FC对人胶质瘤细胞系U251的增殖具有明显的抑制作用,但对原癌基因Fas/FasL的表达无影响。
Objective To investigate the inhibitory effect of C (FC) on the glioma U251 cell line and the expression of the oncogene Fas / FasL. Methods MTT assay was used to detect the proliferation of U251 cells treated with different concentrations of FC. The expression of Fas / FasL was observed by immunohistochemistry and RT-PCR. Results With the increase of FC concentration, the inhibition of proliferation of U251 cells gradually increased in a dose-dependent manner. The results of immunohistochemistry and RT-PCR showed that the expression of Fas / FasL did not change significantly with the increase of FC dosage. Conclusion FC inhibited the proliferation of human glioma cell line U251, but had no effect on the expression of Fas / FasL.
