采用PCR技术对江西瑞昌、湖南长沙、天津宁河3个群体的144尾草鱼线粒体DNA(m tDNA)D-Loop区段的限制性片段长度多态性(RFLP)进行了分析。PCR扩增出的m tDNA D-Loop区段,用11种核酸内切限制酶酶切。结果显示:扩增出的142尾试验鱼的m tDNA D-Loop区段为1.6 kbp,长沙群体中的两尾为1.8kbp,个体间存在长度变异现象;6种限制酶具有酶切位点,共检测到两种单倍型,其中长沙群体中有长度变异的两尾为一种单倍型,142尾为另一种单倍型;瑞昌和宁河两群体内的单倍型多样性指数、核苷酸多样性指数均为0,长沙群体内的分别为0.0816、0.00315;长沙群体与瑞昌(或宁河)群体间的核苷酸歧化距离以及Rogers遗传距离分别为0.0000343和0.6783;χ2检验结果显示3个群体间的遗传差异不显著(P>0.05)。表明离草鱼m tDNA D-Loop区段的遗传多样性很低。
The restriction fragment length polymorphism (RFLP) of mitochondrial DNA (m tDNA) D-Loop segment of 144 Carassius aurats from 3 populations in Ruichang of Jiangxi, Changsha of Hunan and Ninghe of Tianjin was analyzed by PCR. The mtDNA D-Loop segment amplified by PCR was digested with 11 endonucleases. The results showed that the mtDNA D-Loop segment of the amplified 142 fish was 1.6 kbp, and the length of the two populations was 1.8 kbp in Changsha population. There were 6 restriction enzymes with restriction sites, Two haplotypes were detected, of which two mutants in Changsha population had one haplotype and the other 142 haplotypes. The haplotype diversity index in both Ruichang and Ninghe populations , The nucleotide diversity index was 0, and the populations in Changsha were 0.0816 and 0.00315, respectively. Nucleotide disproportionation distance and Rogers distance between Changsha population and Ruichang (or Ninghe) population were 0.0000343 and 0.6783, respectively; χ2 The test results showed that the genetic differences among the three groups were not significant (P> 0.05). Indicating that the genetic diversity of mtDNA D-Loop segments from grass carp is very low.
