用干冰(CO_2)处理B(W_2/W_2)和Cre(re/re)正反交配的初期发生卵,能够诱发精核、卵核及嵌合发生卵,进行了如下几方面的研究。 1.调查了产卵后的经过时间和精核、卵核及嵌合发生卵率的关系。结果认为精核发生卵率以产卵后70~75分钟处理区多,B×Cre达70%,Cre×B为48%。卵核及嵌合发生卵率以产卵后60~70分钟最高,但比精核发生卵率低。 2.产卵后70分钟的蚕卵封入干冰(CO_2)中不同时间,认为B×Cre90分钟处理区的精核发生卵率最高达68.4%;Cre×B150分钟处理区精核发生卵率最高为36%。 3.精核及卵核发生卵的发生调查结果:精核发生卵的孵化率仅为1.6%;化乌死卵70%,解剖致死卵确认了胚子的致死时期。 4.用纸上色层分析法分析卵的色素确认,干冰(CO_2)处理诱发的精核、卵核发生卵的色素是re和W_2基因决定的。 5.探讨了干冰(CO_2)处理,诱发精核、卵核及嵌合发生卵的机理,认为在卵内引起CO_2+H_2OH_1CO_3的反应,使卵内pH下降而诱发的可能性。
Treatment of the initial egg produced by reciprocal mating of B (W 2 / W 2) and Cre (re / re) with dry ice (CO 2) can induce the nucleus, egg nucleus and chimeric egg formation. The following aspects were studied. 1. Investigated the relationship between the elapsed time after spawning and the spermatogenesis, egg nucleus and chimerism. The results showed that the rate of egg occurrence in sperm nucleus was higher in 70-75 minutes after spawning, with B × Cre up to 70% and Cre × B up to 48%. Eggs and chimeric eggs occur in order to spawn 60 to 70 minutes after the highest, but lower than the incidence of fine nucleus eggs. The silkworm eggs at 70 minutes after spawning were filled in dry ice (CO 2) at different times, and the highest incidence of spermatogenesis was 68.4% in the treatment of B × Cre for 90 minutes. The highest incidence of spermatogenesis in the treatment of Cre × B150 was 36%. 3. The results of the occurrence of spermatogenesis and egg ova The results showed that the rate of hatching of the egg produced by sperm nucleus was only 1.6%; the death egg was 70% and the lethal egg was dissected to confirm the lethal period of the embryo. Analysis of egg color by paper chromatography analysis confirmed that the nucleus caused by dry ice (CO_2) treatment was determined by re and W_2 genes. The mechanism of CO 2 treatment, induced nucleus, egg nucleus and chimeric egg development was discussed. It is considered that the reaction of CO 2 + H_2O_H_ 1 CO_ 3 in eggs causes the decrease of pH in eggs.
