[目的]研究6℃低温处理对沼气低温发酵微生物菌群产气特性及菌群数量变化的影响。[方法]利用生化培养箱控制温度条件,采用批量发酵、排水集气和MPN计数法,了解6℃条件沼气发酵器中的主要微生物菌群的数量。[结果]结果表明:通过6℃低温驯化,沼气发酵微生物菌群能维持日平均产气量为23.6 ml/2 500 ml发酵液;发酵细菌数量为3×108个/ml,纤维素分解菌为9×104个/ml;硫酸盐还原菌数量为1.9×105个/ml,其种群数量与常温条件相近;产甲烷菌数量为4×105个/ml,比常温条件低2个数量级。[结论]发酵体系中,低温主要影响产甲烷菌群。
[Objective] The research aimed to study the effects of 6 ℃ low temperature treatment on the gas production characteristics and the change of the number of microbes in biogas fermentation at low temperature. [Method] The temperature of biochemical incubator was used to control the temperature conditions. The batch fermentation, drainage gas collection and MPN counting method were used to understand the main microbial flora in biogas fermentor at 6 ℃. [Result] The results showed that the biogas fermentation microbial flora could maintain the average daily gas production of 23.6 ml / 2 500 ml by acclimation at 6 ℃. The number of fermentative bacteria was 3 × 108 cells / ml and the cellulolytic bacteria was 9 × 104 / ml. The number of sulfate-reducing bacteria was 1.9 × 10 5 / ml. The number of its population was similar to that at room temperature. The number of methanogens was 4 × 105 / ml, which was 2 orders of magnitude lower than that of normal temperature. [Conclusion] In fermentation system, low temperature mainly affected methanogens.
