目的:观察健脾消癌方及其拆方对肠癌模型裸鼠体重、瘤体、转移率及半胱氨酸天冬氨酸蛋白酶-3(Caspase-3),抗凋亡蛋白B细胞淋巴瘤/白血病-xL(B-cell lymphoma/leukemia-xL,Bcl-xL)和Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)表达的影响。方法:建立裸鼠肠癌原位瘤模型,分为模型组,健脾益气组(15 g·kg~(-1)),化瘀解毒组(15 g·kg~(-1)),健脾消癌方组(全方组,15 g·kg~(-1)),5-氟尿嘧啶组(5-Fu,20 mg·kg~(-1)),共5组。分别予以相应药物干预4周,观察裸鼠体重、瘤体抑制率、转移率,蛋白免疫印迹法(Western blot)检测Caspase-3,Bcl-xL,Bax蛋白表达。结果:健脾益气组、全方组体重下降低于模型组(P<0.05),化瘀解毒组体重下降高于模型组(P<0.05);健脾益气组对瘤体的抑制与模型组比较无统计学差异,与空白组比较,化瘀解毒组、全方组对瘤体均有较好抑制作用(P<0.05);各组均出现不同程度地转移,其中模型组、健脾益气组总体转移率达90%,化瘀解毒组、全方组转移率分别为70%,60%,低于模型组(P<0.05);与模型组比较,化瘀解毒组及全方组能够上调Bax蛋白表达,下调Caspase-3,Bcl-xL蛋白表达(P<0.01)。结论:健脾消癌方中健脾益气组优势在于稳定体重,化瘀解毒组优势在于抑制肿瘤,抑制转移;全方组能够稳定体重,抑制肿瘤,抑制转移,全方配伍疗效最优,其抗肿瘤机制可能是上调凋亡相关蛋白Bax及下调Caspase-3,Bcl-xL,诱导肿瘤细胞调亡。
Objective: To observe the effects of Jianpi Xiaohua Decoction and its decomposed formulas on body weight, tumor size, metastasis rate and the expressions of caspase-3, anti-apoptotic protein B cell lymphatic (Bcl-2, Bcl-2, Bcl-2, Bcl-2 and Bcl-2) Methods: The model of orthotopic colon cancer in nude mice was established and divided into three groups: model group, Jianpi Yiqi group (15 g · kg -1), Huayu Jiedu group (15 g · kg -1) Jianpi Xiaowanfang group (15g · kg -1) and 5-Fu (20 mg · kg -1) in 5 groups. The corresponding drug intervention was given for 4 weeks. The body weight, tumor inhibition rate, metastasis rate and protein expression of Caspase-3, Bcl-xL and Bax were detected by Western blot. Results: The body weight of Jianpi Yiqi group and Quanfang group were lower than that of model group (P <0.05), while the body weight of Huayujiedu group was lower than that of model group (P <0.05) Compared with the blank group, Huayu detoxification group and Quan Fang group had better inhibitory effect on the tumor (P <0.05), and all the groups showed different degrees of metastasis, in which model group, healthy group The total transfer rate of spleen and qi group was 90%, and the metastasis rate of Huayu detoxification group and whole group was 70% and 60%, respectively, lower than that of the model group (P <0.05). Compared with the model group, The square group was able to up-regulate Bax protein expression and down-regulate Caspase-3, Bcl-xL protein expression (P <0.01). Conclusion: Jianpi Xiaowan Fang Jianpi Yiqi group has the advantage of stable body weight, blood stasis detoxification group is the advantage of inhibiting tumors, inhibiting metastasis; whole group can stabilize the body weight, inhibit the tumor, inhibit metastasis, the best effect of the whole compatibility, the anti- Tumor mechanism may be upregulation of apoptosis-related protein Bax and downregulation of Caspase-3, Bcl-xL, induce apoptosis of tumor cells.
