目的:考察碱性成纤维细胞生长因子(bFGF)对局部脑缺血损伤和抗氧酶活性的影响.方法:大鼠用插丝法阻塞大脑中动脉24h,梗塞灶用TTC染色显示,图象分析测量;神经功能缺损采用0-5级评分.分别分析每只大鼠左右两侧大脑的4种抗氧化酶活性水平.结果:bFGF(缺血5min后以45μg·kg-1·h-1给予3h)可降低梗塞灶体积和神经功能缺损评分.局部脑缺血升高过氧化氢酶(CAT),超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSHPX)的活性,但使谷胱甘肽S转移酶(GSHST)的活性水平降低.bFGF可在缺血状态下进一步升高CAT的活性,但对SOD,GSHPX和GSHST的水平没有影响.结论:bFGF对局部脑缺血损伤具有神经保护作用,此作用可能与CAT活性水平的升高有关.
Objective: To investigate the effect of basic fibroblast growth factor (bFGF) on the focal cerebral ischemia injury and the antioxidant enzyme activity. Methods: The middle cerebral artery occlusion was occluded for 24 h in rats by plugging method. TTC staining was performed on the infarct area and image analysis was performed. The neurological deficits were assessed by 0-5 scale. The levels of 4 antioxidant enzymes in the left and right brains of each rat were analyzed. Results: bFGF (given at 45 μg · kg-1 · h-1 for 5 h after 5 min of ischemia) decreased infarct volume and neurological deficit scores. Local cerebral ischemia increased catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) activity, but the glutathione S transferase ( GSH ST) activity level decreased. bFGF can further increase the activity of CAT in the ischemic state, but has no effect on the levels of SOD, GSH-PX and GSH-ST. CONCLUSION: bFGF has neuroprotective effect on focal cerebral ischemia injury, which may be related to the increase of CAT activity.
