为了探讨Bcl 2反义核酸治疗Burkitt’s淋巴瘤 (BL)的疗效 ,将不含EB病毒的人类BL的细胞株—CA4 6移植到BAB C裸鼠体内 ,建立BL裸鼠模型 ,并将荷瘤裸鼠随机分为反义治疗 (A组 )、无义对照 (B组 )和空白对照 (C组 ) 3个组 ,每组 5只。A组注射Bcl 2反义核酸 ,B组注射无义寡核苷酸 ,C组不注射任何制剂 ;治疗剂量为 5mg·kg-1 ·d-1 ,给药方式为瘤内局部注射 ,每天注射 1次 ,连续给药 1 5d。结果显示 :A组肿瘤的生长明显受到抑制 ,其中 1只裸鼠的肿瘤消失 ,其余 4只的瘤块明显小于B组和C组。A组对C组的肿瘤抑制率为 89.5 % ;A组对B组的肿瘤抑制率为83.3% ,2者的抑制率无显著差异 (p >0 .0 5 ) ;而B组对C组的抑制率为 36 .9%与前 2者的抑制率差异非常显著 (p <0 .0 1 )。说明Bcl 2反义核酸在体内可以特异性地抑制BL细胞的生长
To investigate the efficacy of Bcl 2 antisense oligonucleotide in the treatment of Burkitt’s lymphoma (BL), human EB cell line-CA4 6 without Epstein-Barr virus (EBV) was transplanted into BAB C nude mice to establish a nude mice model of BL. The rats were randomly divided into three groups: antisense treatment (group A), nonsense control (group B) and blank control (group C). In group A, Bcl 2 antisense oligonucleotide was injected, group B was injected with non-sense oligonucleotide, and group C was not injected with any agent. The treatment dose was 5 mg · kg -1 · d -1. 1 times, continuous administration of 15d. The results showed that the growth of tumor in group A was obviously inhibited, and the tumor in one of the nude mice disappeared. The tumor mass in the other four was significantly smaller than those in group B and C. The tumor inhibition rate of group A was 89.5%, that of group A was 83.3% and that of group B was no significant difference (p> 0.05), while that of group B was 89.5% The inhibition rate of 36.9% was significantly different from the former two (p <0.01). Bcl 2 antisense nucleic acid can specifically inhibit the growth of BL cells in vivo
