本文报道以一例女性肺癌患者胸积液经体外培养而建成的细胞系,定名为PC46。其细胞形态以多核性上皮样细胞为主,在电镜下观察,细胞表面具有致密微绒毛,细胞核形态不规则,核膜有明显凹陷,形成核内假包涵体。细胞的染色体数目范围为60~180,大多数细胞为近三倍体。经G带分析,发现其中有三种标记染色体,并具有双着丝粒染色体、双微体和染色体断片。本细胞群体倍增时间为23小时,分裂指数为207‰,能被Con,A发生凝集。异种移植能生长瘤结节,瘤细胞类似低分化腺癌的组织结构,PAS染色阳性反应。本细胞系的生物学性质,表明它保留了癌细胞的特性。
This article reports a case of female lung cancer patients with thoracic fluid cultured in vitro cell line, named PC46. The morphology of the cells was mainly multinucleated epithelioid cells. Observed under electron microscope, the cells had dense microvilli on the surface, irregular nuclear morphology, obvious depression of the nuclear membrane, and formation of pseudonuclear inclusions in the nucleus. The number of chromosomes in the cells ranged from 60 to 180, with most cells nearly triploid. By G-band analysis, found that there are three markers chromosomes, and with dicentric chromosomes, mitochondria and chromosome fragments. The doubling time of this cell population was 23 hours, and the division index was 207 ‰, which could be condensed by Con and A. Xenografts can grow tumor nodules, tumor cells similar to poorly differentiated adenocarcinoma of the organizational structure, PAS staining positive reaction. The biological properties of this cell line indicate that it retains the characteristics of cancer cells.
