药物疗效和毒性存在个体差异是临床常见的现象,癌症患者接受化疗时更为明显,一直受到临床医生关注。不少临床因素影响人体对药物的反应,但是随着分子生物学、分子遗传学与分子药理学,特别是基因组学的发展,人们逐渐认识到不同个体对同一药物的不同反应大多源于基因的多态性。文章总结二氢嘧啶脱氢酶及其基因单核苷酸多态性的研究进展,理解两者的关系有利于更好地实现5-氟尿嘧啶个体化化疗。
Individual differences in the efficacy and toxicity of drugs is a common clinical phenomenon, cancer patients receiving chemotherapy is more obvious, has been the concern of clinicians. Many clinical factors affect the body’s response to drugs, but with the development of molecular biology, molecular genetics and molecular pharmacology, especially genomics, people gradually come to realize that the different reactions of different individuals to the same drug mostly come from the gene Polymorphism. The article summarizes the research progress of dihydropyrimidine dehydrogenase and its gene single nucleotide polymorphism, understand the relationship between the two is conducive to better individualized 5-fluorouracil chemotherapy.
