目的:研究人乳头瘤病毒16型(HPV16)E7蛋白阳性(E7+)和阴性(E7-)乳癌组织中细胞周期调控相关基因蛋白表达的变化。方法:56例乳癌组织经PCR筛选证实HPV16 DNA阳性,其中42例HPV16 E7+,14例E7-。采用Western blotting方法检测癌组织中细胞周期调控相关基因蛋白CyclinA、CyclinD、CyclinE、P21和P27的表达。结果:42例HPV16E7+乳癌组织中,CyclinA、CyclinD、CyclinE、P21和P27蛋白阳性率分别为97.6%(41/42)、76.2%(32/42)、92.9%(39/42)、9.5%(4/42)和40.5%(17/42);14例HPV16E7-乳癌组织中,CyclinA、CyclinD、CyclinE、P21和P27蛋白阳性率分别为64.3%(9/14)、78.6%(11/14)、57.1%(8/14)、50.0%(7/14)和42.9%(6/14)。与HPV16E7-乳癌组织相比,HPV16E7+乳癌组织中CyclinA和CyclinE蛋白表达均上调(χ2分别为12.196,9.929,P均<0.01),P21表达下调(χ2=10.898,P<0.01),而CyclinD和P27的表达均无明显变化(χ2分别为0.033,0.025,P均>0.05)。结论:HPV16E7可通过统计量上调CyclinA和CyclinE表达以及下调P21表达而在乳癌发生发展中发挥重要作用。
Objective: To study the changes of cell cycle related gene protein expression in human papillomavirus type 16 (HPV16) E7 positive (E7) and negative (E7-) breast cancer. Methods: 56 cases of breast cancer tissues were confirmed by PCR screening HPV16 DNA positive, of which 42 cases of HPV16 E7 +, 14 cases of E7-. Western blotting was used to detect the expression of CyclinA, CyclinD, CyclinE, P21 and P27 in cancer tissues. Results: The positive rates of CyclinA, CyclinD, CyclinE, P21 and P27 in 42 cases of HPV16E7 + breast cancer were 97.6% (41/42), 76.2% (32/42), 92.9% (39/42) and 9.5% The positive rates of CyclinA, CyclinD, CyclinE, P21 and P27 in 14 cases of HPV16E7-breast cancer tissues were 64.3% (9/14) and 78.6% (11/14), respectively , 57.1% (8/14), 50.0% (7/14) and 42.9% (6/14). Compared with HPV16E7-breast cancer, the expression of CyclinA and CyclinE in HPV16E7 + breast cancer were both up-regulated (χ2 = 12.196,9.929, P <0.01) and P21 was down-regulated (χ2 = 10.898, P <0.01) (Χ2 = 0.033, 0.025, P> 0.05). Conclusion: HPV16E7 plays an important role in the development of breast cancer by up-regulating the expression of CyclinA and CyclinE and down-regulating the expression of P21.
