目的:研究卡介苗细胞壁骨架(BCGCWS)加白细胞介素2(IL2)膀胱灌注对肿瘤组织端粒酶活性的影响,从而探讨该治疗的作用机理。方法:采用端粒重复序列扩增法(TRAP)分别检测20例膀胱肿瘤用药前后端粒酶活性的表达。结果:经分别测定端粒酶提取原液以及10倍、100倍稀释液中端粒酶活性的表达,表明用药后肿瘤组织中端粒酶的活性明显降低(P<001)。结论:BCGCWS联合IL2膀胱灌注能有效地抑制肿瘤细胞端粒酶的活性,提示该治疗的作用机理之一可能是肿瘤端粒酶活性的改变。
Objective: To study the effect of BCGCWS combined with interleukin2 (IL2) intravesical instillation on telomerase activity in tumor tissue and to explore the mechanism of action of this treatment. Methods: Telomerase activity was detected by telomeric repeat amplification (TRAP) in 20 cases of bladder cancer before and after treatment. Results: The telomerase activity and the telomerase activity in the 10-fold and 100-fold dilutions were determined respectively. The results showed that telomerase activity in the tumor tissue was significantly decreased (P <001). Conclusion: BCGCWS combined with IL2 intravesical instillation can effectively inhibit the activity of telomerase in tumor cells, suggesting that one of the mechanisms of action might be the alteration of telomerase activity.
