目的 本文旨在探讨大鼠鼻黏膜P物质受体 (SPR)mRNA的表达水平及其与变应性鼻炎的关系。方法 选健康Wistar大鼠 2 0只 ,雌雄不限 ,随机分为实验组和对照组各 10只。实验组用卵清蛋白行腹腔注射基础致敏 ,继之鼻局部激发建立大鼠变应性鼻炎模型。以 β 肌动蛋白 (β actin)作内标准 ,利用反转录 多聚酶链反应(RT PCR) ,对变应性鼻炎模型组和对照组大鼠鼻黏膜SP RmRNA进行了定量研究。同时利用地高辛标记的cDNA探针对其行Southern杂交鉴定。结果 对照组和实验组大鼠鼻黏膜中均有SP RmRNA表达 ,但实验组大鼠中SPR表达量较对照组明显增多 (P <0 .0 5 )。结论 变应性鼻炎鼻黏膜SP RmRNA表达量增多 ,提示SP RmRNA表达上调在变应性鼻炎的发生发展中起重要作用
Objective To investigate the expression of substance P receptor (SPR) in rat nasal mucosa and its relationship with allergic rhinitis. Methods Twenty healthy Wistar rats were randomly divided into experimental group and control group (n = 10). Experimental group with ovalbumin peritoneal injection of basic sensitization, followed by local nasal stimulation rat allergic rhinitis model. Using β-actin as the internal standard, RT-PCR was used to quantitatively study the nasal mucosa SP RmRNA of allergic rhinitis model group and control group. Southern blotting was also performed using digoxigenin-labeled cDNA probes. Results The expression of SP R mRNA in nasal mucosa of both control and experimental groups was significantly higher than that of the control group (P <0.05). Conclusion The expression of SP R mRNA in nasal mucosa of allergic rhinitis increased, suggesting that the upregulation of SP R mRNA may play an important role in the development of allergic rhinitis
