目的:比较诱导分化剂苯乙酸(PA)及二甲基甲酰胺(DMF)对胶质瘤细胞C6的增殖抑制作用,观察两者对Wistar荷瘤大鼠生存时间的影响,并从作用机理角度探讨两者联合应用的前景。方法:3H-TdR掺入法检测PA及DMF对体外培养的胶质瘤细胞的增殖抑制率;免疫组化SP法检测c-myc、p16基因在蛋白水平的表达。对Wistar荷瘤大鼠进行PA及DMF腹腔注射治疗,观察生存时间变化。结果:应用PA及DMF后,C6细胞CPM值降低;PA组c-myc基因胞浆阳性表达显著下降;DMF组p16基因胞浆阳性表达显著增强。Wistar荷瘤大鼠腹腔注药后,生存时间显著延长。结论:PA及DMF对胶质瘤细胞的增殖在体内外实验中均存在剂量依赖性抑制作用,作用机理显示两者有联合应用的前景。
OBJECTIVE: To compare the effects of phenacetic acid (PA) and dimethylformamide (DMF) on the proliferation of glioma cells C6, and to observe the effects of the two agents on the survival time of Wistar-bearing mice, and from the perspective of the mechanism of action. Explore the prospect of a joint application of the two. METHODS: The inhibition rate of PA and DMF on the proliferation of glioma cells cultured in vitro was detected by 3H-TdR incorporation method. The expression of c-myc and p16 at the protein level was detected by immunohistochemistry SP method. Wistar tumor-bearing rats were treated with intraperitoneal injection of PA and DMF, and the survival time was observed. Results: After the application of PA and DMF, the CPM value of C6 cells decreased; the cytoplasmic positive expression of c-myc gene in PA group decreased significantly; the cytoplasmic positive expression of p16 gene in DMF group increased significantly. After Wistar tumor-bearing rats were injected intraperitoneally, the survival time was significantly prolonged. CONCLUSION: Both PA and DMF have a dose-dependent inhibitory effect on the proliferation of glioma cells in vitro and in vivo.
