目的 以新西兰家兔面神经切断后硅胶管神经再生腔模型 ,研究肝细胞生长因子 (Hepatocytegrowthfactor ,HGF)对面神经损伤后再生修复作用的影响 ,为临床寻找方便、有效的神经营养因子提供初步的实验依据。方法 30只新西兰家兔 ,双侧面神经上颊支横断后用显微外科技术将硅胶管与神经外膜缝合以形成硅胶神经再生腔 ,一侧注入HGF为实验侧 ,另一侧注入生理盐水为对照侧 ,于术后 1、2、4周分别麻醉家兔后行电生理检测 ,在特殊染色下进行组织形态学观察 ,并进行形态定量分析。结果 术后 1周 ,实验侧与对照侧均未见再生的有髓神经纤维。术后 2周 ,两组间即出现差异。术后 4周 ,两组差异有显著性意义。结论 外源性追加HGF对神经再生短期内有促进作用。
OBJECTIVE: To study the effect of Hepatocyte growth factor (HGF) on the regenerative repair after facial nerve injury in New Zealand rabbit model of facial nerve regeneration after facial nerve transection to provide a preliminary experimental evidence for finding a convenient and effective neurotrophic factor . Methods Thirty New Zealand rabbits were transected with buccal branch of the lateral facial nerve by microsurgical technique. The silicone tube and the epineurium were sutured to form a silicone nerve regeneration chamber. HGF was injected into the experimental side on one side and saline was injected on the other side On the control side, rabbits were anesthetized respectively at 1, 2 and 4 weeks after operation, and electrophysiological examination was performed. Morphological observation was performed under special staining and morphometric analysis was performed. Results One week after operation, no regenerated myelinated nerve fibers were found on the experimental side and the control side. Two weeks after surgery, there were differences between the two groups. After 4 weeks, there was significant difference between the two groups. Conclusion Exogenously added HGF can promote short-term neuroregeneration.
