目的:研究苦参碱对人肝癌细胞系体外诱导分化作用,并探讨其作用机制。方法:不同浓度苦参碱体外作用于Bel-7404细胞,噻唑蓝比色法(MTT法)绘制生长曲线,光镜下观察细胞形态变化,分别以软琼脂集落形成试验、细胞黏附试验、运动试验、侵袭试验检测细胞生物学特性变化,以流式细胞仪分析细胞周期变化。结果:0.5-1.0g/L苦参碱作用于Bel-7404细胞后,明显抑制细胞增殖,细胞形态向正常转化,克隆形成率显著降低,细胞黏附力明显上升,运动力和侵袭力则显著下降。细胞周期分析G1期细胞堆积,G2和S期细胞显著降低。结论:一定浓度的苦参碱具有抑制Bel-7404细胞生长增殖和诱导分化作用,其作用机制可能与细胞周期调节有关。
OBJECTIVE: To study the induction and differentiation of matrine on human hepatocellular carcinoma cell line in vitro and its mechanism. METHODS: Bel-7404 cells were treated with different concentrations of matrine in vitro. The growth curve was drawn by MTT assay. The morphological changes of cells were observed under light microscope. Soft agar colony formation assay, cell adhesion assay, , Invasion test to detect changes in cell biological characteristics, flow cytometry analysis of cell cycle changes. Results: After treated with 0.5-1.0 g / L matrine, the proliferation of Bel-7404 cells was significantly inhibited. The cell morphology was transformed into normal cells. The colony formation rate was significantly decreased. The cell adhesion was significantly increased. The motility and invasiveness decreased significantly . Cell cycle analysis G1 phase cell accumulation, G2 and S phase cells decreased significantly. Conclusion: Matrine at a certain concentration can inhibit the growth, proliferation and differentiation of Bel-7404 cells, which may be related to the regulation of cell cycle.
