目的 探讨蛋膜蛋白提取物(Esm)补充对一次性下坡跑运动损伤的影响及可能机制.方法 雄性SD大鼠14只,随机分为普通鼠料对照组(CON)和Esm实验组.Esm实验组每天灌胃补充Esm 200 mg/(kg bw);对照组补充等量的酪蛋白,2w后进行一次性下坡跑运动.运动后空腹12h取材,检测血清乳酸、尿素、丙二醛等指标,肌肉组织检测葡萄糖醛酸酶活性和肿瘤坏死因子α(TNF-α)蛋白表达.结果 一次力竭性运动后,Esm组大鼠血清乳酸、尿素和丙二醛(MDA)浓度均显著低于对照组(P0.05).Esm补充显著降低了骨骼肌组织中葡萄糖醛酸酶活性(P<0.05).蛋白印迹结果显示2w的Esm补充显著降低了运动后骨骼肌组织中TNF-α 蛋白的表达(P<0.05);且比目鱼肌组织中TNF-α 表达显著高于股四头肌组织中的表达(P<0.05).结论 Esm对运动损伤有一定保护作用,其机制可能促进机体自由基清除和调节骨骼肌TNF-α 的表达有关,从而加快运动后疲劳的消除.“,”Objective To investigate the effect of egg membrane (Esm) protein on the injury induced by downhill running and its possible mechanism.Methods Male Sprague-Dawley rats (n=14) were randomly divided into control group (CON) and Esm experimental group. The Esm group was fed by gavage with Esm 200 mg/kg bw every day, and the control group was supplemented with the same dose of casein. A one-time downhill running test was carried out after two weeks of supplementation. After fasting for 12h, samples were collected. Serum lactic acid, urea and malondialdehyde (MDA) were detected, and the changes of glucuronid activity and the expression of TNF-α protein was detected in skeletal muscles. Results After the downhill running, the levels of serum lactic acid, urea and MDA were significantly lower in Esm group than those in the control group (P<0.05). The serum creatine kinase (CK) and lactate dehydrogenase (LDH) in Esm group were lower than those in control group, but there was no significant difference. The glucuronid activity in soleus and quadriceps muscles was significantly lower than that in the control group (P<0.05). The result of Western blot showed that TNF-α expression in soleus muscle tissue was significantly higher than that in quadriceps muscle (P<0.05); and Esm supplementation significantly reduced the expression of TNF-α in skeletal muscles after the downhill running (P<0.05). Conclusion Egg membrane protein has a certain protective effect on sports injury, and the mechanism is related to the scavenging of free radicals and decreased expression of TNF-α in skeletal muscles.
