我们制备了上海,广西和郑州三株中华按蚊,江苏和广西两株嗜人按蚊的基因组DNA,分别用BglⅡ,HaeⅢ和PstⅠ消化各基因组DNA,进行琼脂糖凝胶电泳再用溴乙锭染色后,观察比较同种按蚊各株间基因组DNA重复序列DNA的限制酶切长度差异.结果发现,各地理株间的酶切带型,既具有种的特征,即主带相同,稳定;又有株的特异性淡带。三株中华按蚊的PstⅠ,两株嗜人按蚊的BglⅡ的酶切长度差异可以将各地理株区别开来,比较观察限制酶切长度差异是分析种内各地理株变异的敏感可靠的方法.实验结果表明了中华按故三地理株,嗜人按蚊的两地理株间的重复序列DNA的结构有差异,从基因结构上证实了两种按蚊种下的分化。
We prepared genomic DNA of three Anopheles anthropophagus, Anopheles sinensis, Jiangsu and Guangxi strains in Shanghai, Guangxi and Zhengzhou.
