目的研究四君子汤对结肠腺癌荷瘤小鼠癌性恶病质的影响及可能作用机制。方法 30只Balb/c小鼠随机分为正常组、模型组、四君子汤组,每组10只。模型组和四君子汤组予右侧前背部皮下注射结肠腺癌CT26细胞约1×107个,皮下种植肿瘤细胞后第1天开始模型组和正常组灌胃0.2ml生理盐水,四君子汤组灌胃0.2ml四君子汤药液,相当于3.01g/(kg·d)药粉,各组均灌胃15天。记录不同时间小鼠体重、饮食总量、平均每天饮食量,16天后取材比较脏器重量、瘤重及生化指标、炎症因子白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)含量。结果与正常组比较,模型组小鼠在第5天开始饮食摄入总量较正常组与四君子汤组减少,在第11-16天平均进食量亦明显低于四君子汤组及正常组(P<0.05);四君子汤组小鼠体重第16天时较正常组低,但明显高于模型组(P<0.05);模型组与四君子汤组小鼠脏器重量、瘤重、生化指标比较差异无统计学意义(P>0.05);与模型组比较,四君子汤组血清TNF-α、IL-6明显降低(P<0.05)。结论四君子汤能有效减轻结肠腺癌荷瘤小鼠癌性恶病质症状,提高体重,抑制炎症因子TNF-α、IL-6可能其作用机制之一。
Objective To study the effect of Sijunzi Decoction on cancerous cachexia in colon adenocarcinoma mice and its possible mechanism. Methods Thirty Balb / c mice were randomly divided into normal group, model group and Sijunzi Decoction group with 10 rats in each group. The model group and the Sijunzi Decoction group were given 1 × 107 CT26 cells subcutaneously injected into the right anterior dorsal subcuta. After the tumor cells were subcutaneously implanted, the model group and the normal group were given intragastric administration of 0.2 ml normal saline and Sijunzi Decoction 0.2ml Sijunzi decoction, equivalent to 3.01g / (kg · d) powder, each group were fed for 15 days. The body weight, total diet and average daily diet of mice at different time points were recorded. After 16 days, the weight of organ, tumor weight and biochemical indexes, interleukin-6 (IL-6), tumor necrosis factor- -α) content. Results Compared with the normal control group, the dietary intake of the mice in the model group decreased on the 5th day compared with that of the normal group and the Sijunzitang group, and the average food intake on the 11th to 16th day was also significantly lower than that of the Sijunzi Decoction group and the normal group (P <0.05). The body weight of Sijunzi Decoction group was lower than the normal group on the 16th day, but was significantly higher than that of the model group (P <0.05). There was no significant difference in the organ weight, tumor weight and biochemical index between the model group and the Sijunzitang group (P> 0.05). Compared with the model group, the serum levels of TNF-α and IL-6 in Sijunzi Decoction group were significantly decreased (P <0.05). Conclusion Sijunzitang can effectively reduce the symptoms of cancer cachexia in colon adenocarcinoma mice, increase body weight and inhibit the inflammatory cytokines TNF-α, IL-6 may be one of the mechanisms.
