目的:研究大鼠牙髓损伤修复过程中串珠素的时空表达变化及作用机制。方法 :选用24只雄性SD大鼠,通过暴露一侧大鼠上颌磨牙牙髓建立牙髓损伤模型,对侧未损伤磨牙作为对照组,术后1、3、5、7 d取标本,脱钙后石蜡包埋,近远中向沿牙体长轴4μm切片,采用免疫组织化学染色观察串珠素和β1整合素的表达变化,平均吸光度法计算相对表达量,采用SPSS19.0软件包对数据进行单因素方差分析。结果:与对照组相比,实验组串珠素染色强度增加,阳性染色位置随着炎症损伤的进展方向而转移,各组平均光密度均高于对照组(F=9.511,P<0.05),并以1 d组升高最为明显。β1整合素染色在牙髓损伤后3 d开始,在炎症牙髓和相对正常牙髓中阳性表达,3、5、7 d组的平均吸光度均显著高于对照组(F=124.18,P<0.05)。结论:串珠素在牙髓组织损伤后防御机制中可能通过调节血管生成和炎症细胞浸润而发挥作用。
OBJECTIVE: To study the temporal and spatial expression of perlecan and its mechanism during the repair of rat dental pulp injury. Methods: Twenty-four male Sprague-Dawley rats were used to establish the model of dental pulp injury by exposing the dental pulp of the maxillary molars of one side to the untreated molar. The rats were sacrificed at 1, 3, 5 and 7 days after operation, After paraffin embedding, in the near and far along the long axis of the tooth 4μm section, immunohistochemical staining of perlecan and β1 integrin expression changes, the average absorbance method to calculate the relative expression level, the data using SPSS19.0 package ANOVA. Results: Compared with the control group, the streptozotocin staining intensity increased and the positive staining position shifted with the progress of inflammatory injury. The average optical density of each group was higher than that of the control group (F = 9.511, P <0.05) In the 1 d group increased the most obvious. The β1 integrin staining began to be expressed on the 3rd day after dental pulp injury, and was positively expressed in inflammatory pulp and normal dental pulp. The average absorbance in 3, 5, 7 d group was significantly higher than that in control group (F = 124.18, P <0.05 ). CONCLUSION: Perlecan may play a role in the defense mechanism of dental pulp injury by regulating angiogenesis and inflammatory cell infiltration.
