目的探讨胰岛素样生长因子及其I型受体抗体对人肾上腺皮质癌SW-13细胞凋亡的调控和其可能机制。方法原位凋亡(TUNEL)法检测胰岛素样生长因子及其I型受体抗体单独或联合作用于SW-13细胞后细胞的凋亡的情况,同时观察细胞形态及数量的变化。结果各组细胞凋亡率比较有显著统计学差异(P<0.001),其中单用IGF-I组或IGF-II组细胞凋亡率低于空白对照组低于混合作用组低于单用anti-IGF-IR组,而单用IGF-I组和IGF-II组、IGF-I+anti-IGF-IR组和IGF-II+anti-IGF-IR组组间比较无统计学差异(P>0.05)。加入anti-IGF-IR组细胞形态也发生了明显的变化。结论胰岛素样生长因子可促进SW-13细胞株的生长,这与其抑制了细胞凋亡有关,而这种生物学作用是通过胰岛素样生长因子I型受体介导,胰岛素样生长因子I型受体抗体可明显抑制这种生物学作用,可能在肾上腺皮质癌的治疗中发挥重要作用。
Objective To investigate the regulation of insulin-like growth factor and its type I receptor antibody on the apoptosis of human adrenocortical carcinoma SW-13 cells and its possible mechanism. Methods TUNEL method was used to detect the apoptosis of SW-13 cells treated with insulin-like growth factor and its type I receptor antibody alone or in combination. At the same time, the changes of cell morphology and quantity were observed. Results The apoptotic rates in each group were significantly different (P <0.001). The apoptotic rates in IGF-I group and IGF-II group were lower than those in the blank control group IGF-I + anti-IGF-IR group and IGF-II + anti-IGF-IR group had no significant difference (P> 0.05). Addition of anti-IGF-IR group cells also changed significantly. Conclusion Insulin-like growth factor (IGF-1) can promote the growth of SW-13 cell line, which is related to the inhibition of apoptosis. This biological effect is mediated by insulin-like growth factor type I receptor. Insulin-like growth factor type I receptor Antibodies can significantly inhibit this biological role, may play an important role in the treatment of adrenal cortical cancer.
