背景:神经干细胞在神经系统退行性疾病中的疗效日益得到肯定,但人不同胚龄神经干细胞的发育规律评估较少。目的:探讨不同胎龄人纹状体神经干细胞的发育规律,为拓展神经干细胞在儿科领域的应用打下基础。设计:非随机对照的实验研究。地点、材料和干预:本研究在全军儿科中心实验室进行。30例水囊引产的胎儿经家属及胎儿母亲同意后由广州市某三级甲等医院妇产科提供,并按胎龄24,26,28,30,32周分为5组,每组6例。采用免疫组织化学和光镜观察技术进行实验。主要观测指标:人不同胎脑纹状体神经干细胞的形态及其生长方式。结果:不同胎龄纹状体组织均存在神经干细胞,细胞呈圆形、椭圆形及三角形,以圆形和椭圆形多见,三角形细胞仅出现在30,24周的胎脑纹状体。各型细胞有大有小,0~3个突起,核呈圆形及椭圆形,大部分细胞染色质疏松,少部分细胞染色质致密,1~4个核仁不等。5个胎龄的大多数神经干细胞以单个细胞形式生长;30周神经干细胞偶见对称性分裂的集落样生长方式;28周神经干细胞可见对称分裂双细胞、多个细胞集落等生长方式。同胎龄纹状体神经干细胞的形态、大小及生长方式基本相同。结论:人胎脑纹状体存在神经干细胞,且不同胎龄纹状体的神经干细胞在形态及生长方式上存在一定的差异。
Background: The efficacy of neural stem cells in neurodegenerative diseases is increasingly recognized, but the developmental rules of neural stem cells in different embryonic age are less evaluated. Objective: To investigate the development of neural stem cells in different gestational age human striatum and lay the foundation for expanding the application of neural stem cells in pediatrics. Design: Non-randomized controlled experimental study. Location, Materials and Interventions: This study was conducted in the PLA Pediatric Center Laboratory. Thirty patients with induced abortion of the water sac were provided by the obstetrics and gynecology department of a tertiary level hospital in Guangzhou City with the consent of their relatives and their mothers and were divided into 5 groups according to gestational age of 24, 26, 28, 30, 32 weeks example. Immunohistochemistry and light microscopy were used to carry out experiments. MAIN OUTCOME MEASURES: Morphology and growth patterns of neural stem cells from different human fetal striatum. RESULTS: Neural stem cells were found in striatum of different gestational age. The cells were round, oval and triangular, more common in round and oval shape. Triangular cells only appeared in fetal brain striatum at 30 and 24 weeks. Each type of cells are large and small, 0 to 3 protrusions, the nucleus was round and oval, most of the cells chromatin loose, a small number of cells chromatin dense, ranging from 1 to 4 nucleolus. The majority of neural stem cells of 5 gestational age grew as single cells; the colony-like growth pattern of symmetrical divisions of neural stem cells was found at 30 weeks; and the growth pattern of symmetrical dividing double cells and multiple cell colonies was seen at 28 weeks. With the gestational age striatal neural stem cells morphology, size and growth pattern is basically the same. Conclusion: There are neural stem cells in striatum of human fetus, and there are some differences in morphology and growth pattern of neural stem cells in different gestational age.
