测定细胞内Ca~(2+)浓度已成为研究脑水肿发生机制的焦点。本文采用Fura-2荧光标记法,测定冷冻伤性脑水肿时突触体内游离钙离子浓度变化,来探讨突触体的制备方法、Fura-2测定神经细胞突触体内Ca~(2+)浓度的原理、方法及Fura-2优缺点和其影响因素。
Determination of intracellular Ca ~ (2+) concentration has become the focus of research on the mechanism of brain edema. Fura-2 fluorescence labeling method was used to determine the changes of intracellular free calcium concentration in synaptosomal injury in patients with cold-induced traumatic cerebral edema to explore the preparation of synaptosomes. Fura-2 was used to determine the concentration of Ca2 + in neurons The principle and method and the advantages and disadvantages of Fura-2 and its influential factors.
